CZ BioMed Corp. Announces Successful Reduction of Prostate Cancer Tumors More Than 50% Using PRO-RVLYSIS®

TAMPA, FL--(August 26, 2013) -  CZ BioMed Corp announced today it has received positive results for Atlanta, GA patient, Peter V., diagnosed with advanced prostate cancer with extracapsular disease associated with extensive pelvic lymphadenopathy and osseous metastatic disease. Studies of MRIs, before and after treatment, accurately demonstrate a greater than 50% reduction in tumor volumes.

Peter V. is a 59 year old white male in otherwise healthy condition. The cancer itself is further classified as a Stage 4, hypointense T2 tumor engulfing the entire prostate. The tumor initially was invading the base of the bladder, seminal vesicles and distal sphincter with gross extracapsular extension into the left and right neurovascular bundles. Prior to treatment, a multiparametricMRI was performed by Partners Imaging Center of Sarasota, Florida using a Siemens 3.0 TeslaMRI on 04/01/2013. Soon after, the patient began 3 months of hormone therapy utilizing an LHRH analog ending 07/10/13. Simultaneously in April, Peter was selected for clinical trials overseas, and received 24 doses of CZ Biomed's novel oncolytic virotherapy, PRO-RVLYSIS® (twice a day for 12 days). The PRO-RVLYSIS® treatment was overseen by CZ Biomed's medical director Dr. Ronald Wheeler, M.D. It should be noted that PRO-RVLYSIS® was administered systemically, via I.V. only.

On 07/20/2013 a follow up scan, using the same model MRI, was performed at Emory University Hospital in Atlanta, GA. Partners Imaging Center's Dr. Richard Goldberg, M.D. analyzed and compared the imaging studies. The volume of the primary tumor had decreased by more than 50% since the prior exam from 8.0 x 6.7 x 6.6 cm3 to 6.3 x 5.4 x 4.7 cm3. In addition, there was a reduction in size of the bilateral lower pelvis lymphadenopathy. A large nodal mass within the right lower pelvis has decreased from 6.2 x 4.9 cm to 5.2 x 3.6 cm with less mass effect upon the right lateral urinary bladder. A nodal mass within the left lower pelvis has decreased from 5.2 x 4.1 cm to 3.7 x 2.5 cm. Multiple other lymph nodes have decreased as well. Furthermore, the multiple bony metastases were less apparent in the post treatment images and have faded in signal since the prior exam and the urinary bladder was no longer distended. There is no evidence for progressive or new disease. Finally, a PSA Test (prostate-specific antigens) was also performed before and after treatment. The initial PSA reading was 490 ng/mL and 3 months post-treatment PSA reading was only 67 ng/mL, a marked decrease.

These reductions are no doubt as a result of the two week systemic administration of PRO-RVLYSIS®, as it is well known that the LHRH analog therapy alone only slows the growth of tumors and has not ever characteristically reduced or reversed tumor size. PRO-RVLYSIS® is a genetically Engineered oncolytic virus that selectively targets and destroys cancer cells, whilst leaving surrounding non-malignant cells unharmed/uninfected. The destruction of these cancer cells occurs either through direct lytic rupture via multiple cycles of viral replication or the subsequent induction of apoptosis, also known as programmed cell death. Thus, PRO-RVLYSIS® is vastly safer to use and easier for the body to process, as compared to traditional chemo or radio-therapies of tumors, allowing patients to enjoy a much higher quality of life during treatment. Prostate Cancer Trials Underway Overseas PRO-RVLYSIS(R) CZ BioMed Corp.

TAMPA, FL--(January 25, 2012) -  CZ BioMed Corp announced today the successful collaboration with the Florida based Prostate Cancer Prevention Foundation (PCPF), allowing overseas trials of CZ BioMed's new treatment for prostate cancer, PRO-RVLYSIS®. PCPF has assisted by sponsoring treatments for 15 overseas patients which are already underway as of this writing.

PRO-RVLYSIS® is the invention of CZ BioMed Corp. PRO-RVLYSIS® is a novel, genetically engineered common cold-like virus that selectively kills only the Prostate cancer cells, while leaving the normal and healthy cells within the body unharmed. This is considered an oncolytic virotherapy systemic treatment, administered non-invasively through an I.V., which allows the PRO-RVLYSIS® to proceed on its search and destroy mission for cancer cells and is completely safe.

Traditional treatments combine surgery (removal of the prostate), chemo and radiation therapy, along with hormone blockers. Side effects usually involve incontinence and permanent impotence and/or dysfunction. Complete castration as a result of these treatments failing is a final option. These side effects are terrifying for men, and for many, unacceptable. PRO-RVLYSIS® so far has not shown to have any of these side effects.

The results from the 15 patients currently undergoing treatment will be forthcoming, but appear to be very promising thus far. PRORVLYSIS® specifically kills prostate cancer cells (LNCaP cells), but not normal PCS- 440-010 (Human Primary Prostate Epithelial Cells). PRORVLYSIS® selectively induced cytopathic effect (CPE) in the LNCaP prostate cancer cells, and had a higher viral titer in these tumor cells than in the normal PCS-440-010 cells 24h post-infection..

PRORVLYSIS® was also found to kill PC3 cells when checked 48 hours post-infection. This indicates that PRORVLYSIS® has the potential to treat a broad spectrum of prostate cancers.

Moreover, PRORVLYSIS® may prove to induce less neutralizing antibodies upon in vivo administration, because of its lower antigenesis as compared to other oncolytic viruses; similar to our test results with breast cancer cells treated with our BRVLYSIS®. This would suggest that PRORVLYSIS® can be administered multiple times to allow for maximum effect in eradicating the tumor, without concern for the host body rejecting it or fighting off the treatment.

PRORVLYSIS® specifically localizes to the tumor upon direct injection, and does not spread to other healthy tissues or organs due to its genetic modifications. Thus, this procedure is not harming areas of the body that are not currently in danger of the cancer. Intravenous administration for distant cancers is also possible with its use as a pre-cancer vaccination highly plausible at this point due to the virus' ability to exist, and not infect non-cancerous cells.

PANRVLYSIS® was found to effectively kill a variety of pancreatic cancer cell in vitro (within test glass). Additionally, results have shown that PANRVLYSIS® induces tumor growth regression in vivo (within a living organism) as well. This new product has been well tolerated in all experimental mice to date, with PANRVLYSIS® specifically localizing to the tumor upon its local administration, and has not spread to other tissues or organs.     BRVLYSIS is the invention of CZ BioMed Corp. BRVLYSIS is a genetically modified oncolytic virus that selectively targets and destroys breast cancer cells, whilst leaving surrounding non-malignant cells unharmed. The destruction of these cancer cells occurs either through direct lytic rupture via multiple cycles of viral replication or the subsequent induction of apoptosis, also known as programmed cell death. Thus, BRVLYSIS is vastly safer to use and easier for the body to process, as compared to traditional chemo - or radio-therapies of tumors, allowing patients to enjoy a much higher quality of life during treatment.

    BRVLYSIS preferentially kills breast cancer cells both in vitro and in vivo. MDA-MB-231 breast cancer cells and normal CCD-1059SK (Human normal breast fibroblast) were cultured as indicated by ATCC (American Type Culture Collection) instructions, and then infected with the wild type and BRVLYSIS (MOI=5) versions of the virus respectively. BRVLYSIS selectively induced cytopathic effect (CPE) in the MDA-MB-231 breast cancer cells, and had a higher viral titer in these tumor cells than in the normal CCD-1059SK cells 24h post-infection.

    BRVLYSIS was also found to kill T-47D cells, a p53- cell like the subject MDA-MB-231's, as well as p53+ breast cancer cells such as MCF7. This indicates that BRVLYSIS has the potential to treat a very broad spectrum of breast cancers.

    Moreover, BRVLYSIS induced less neutralizing antibodies upon in vivo administration because of its lower antigenesis as compared to other oncolytic viruses. This suggests that BRVLYSIS can be administered multiple times to allow for maximum effect in eradicating the tumor, without concern for the host body rejecting it or fighting off the treatment.

    In an established human breast cancer nude BALB/c mouse model, BRVLYSIS was locally injected into the tumors three times and the sizes of the tumors were significantly reduced by day 8 versus controls. At day 18 post- BRVLYSIS -injection, the sizes of the tumors almost disappeared and have not shown signs of restoration since.

    BRVLYSIS specifically localizes to the tumor upon direct injection, and does not spread to other tissues or organs. Thus, this procedure is not harming areas of the body that are not currently in danger of the cancer. Intravenous administration for distant cancers is also possible with its use as a pre-cancer vaccination highly plausible at this point due to the virus' ability to exist, and not infect non-cancerous cells.

    Breast cancer is the most common cancer among women, with more than one million new cases identified worldwide each year. An estimated 192,370 patients were newly diagnosed with breast cancer in the United States alone in 2009, and about 40,170 died of the disease. Approximately 24% to 30% of women who have no lymph-node involvement at the time of diagnosis will relapse; the relapse rate for node-positive women is between 50%-60%. The 5-year survival rates for those diagnosed with regional and metastatic disease are 80% and 26%, respectively. Therefore, a safe and effective treatment remains a critical need.
